This was the movie that was a must-see for everyone, regardless of age, religion, ideology, whatever. The movie was a transcending experience like Star Wars (or The Dark Knight!) for a later generation.
And of the films on the list, it's really the only one that represents the height of power of the old studio system.

HIS GIRL FRIDAY has more influence specifically within screwball comedies, but IT HAPPENED ONE NIGHT was a major force in creating that genre in the first place, and has had more lasting repurcussions in the broad sphere of romantic comedies afterward.

And if you're talking 'most influential' it's hard to top a film that actually had a huge affect on the sales of real items in the world. And if the Bugs Bunny influence is as true as some say, that's a further claim for Capra's movie.

I don't even see this as a question, really.

EDIT to add: Between the two I'd probably rather watch FRIDAY, but I'd prefer THE AWFUL TRUTH over that. If you're going to angle for FRIDAY being more influential than Capra's movie, then you might have to go back to THE AWFUL TRUTH, which created the enduring Cary Grant persona that was so well refined in FRIDAY.

AS much as IT HAPPENED ONE NIGHT is a screwball, it's probably the most influential Rom-Com ever made, as it ends with the walls of jerhico coming down, which tends to be the punchline for most romantic comedies, until recently.

I was thinking about The Awful Truth earlier today, but I couldn't quite place it in the pioneering vs. influential debate since it was close to IHON and His Girl Friday.

For the sake of argument about what firmly established the rom-com and all that followed, the easy answer is IHON. Most of what has followed in the last 36 hours has been semantics.
