I wouldn't make the case for "greatest", but Craig Robinson totally crushes his 4 lines in Knocked Up.  That was his breakout role, inasmuch as he's had one.

Tommy Lee Jones' uncle in No Country For Old Men comes to mind. No idea who the actor is, but that scene was shockingly the most resonant part of the film for me.

Yeah, the car ride and the desert are what was throwing me. But yeah, I'm cool with that judgement.

 

Also, on the Almost Famous front, add Kyle Gass in there. Sadly, Untitled disqualifies Jimmy Fallon, but that film still represents the only time I have EVER been impressed with him.

I haven't seen every monster movie ever made, but I'm pretty sure the giant spider in "The Lord of the Rings: Return of the King" is the greatest giant insect in movie history. The spider alone made the movie deserve to win best picture. It was that awesome.
